L'étude des complexes antigène-anticorps par cristallographie aux rayons X a montré que la spécificité d'un anticorps dépend des trois régions hypervariables de l'extrémité aminée des chaînes de chaque bras de la molécule. Baptisé «région déterminant la complémentarité» (CDR en anglais), l'ensemble de ces régions constitue le site fixateur d'antigène, assorti physiquement à l'antigène comme un modèle l'est à son moule.
