Three types of reversible inhibition occur. Competitive, which can be reduced by increasing the substrate concentration; non-competitive, which cannot be so reduced; and excess substrate inhibition caused by the formation of non-productive complexes. Plusieurs interprétations ont été proposées. L'une considère que la fixation de substrat supplémentaire dans un second site actif de l'enzyme conduit à une modification de conformation de la protéine, inhibitrice à l'égard du site actif principal. L'autre, peut-être plus réaliste, considère qu'en présence d'un excès de substrat, deux ou plusieurs molécules de ce substrat ont accès au site actif, mais ne contractent avec lui que des liaisons partielles, ce qui rend la catalyse inopérante.
