A yeast autonomously replicating sequence, ARS305, shares essential components with a chromosome 111 replicator, OR1305. Known components include an ARS consensus sequence (ACS) element, presumed to bind the origin recognition complex (ORC), and a broad 3'-flanking sequence which contains a DNA unwinding element.

Clonage de grands fragments d'ADN sous forme de chromosomes artificiels de levure (YAC, yeast artificial chromosomes). Le principe de ce système à munir de grands fragments d'ADN exogène (humain par exemple) de tous les éléments qui permettent leur propagation dans la levure : centromère (CEN), origine de réplication (ARS pour Autonomously Replicating Sequence), telomères (TEL). Ces éléments préalablement clonés à partir de la levure ou de Tetrahymena sont produits - associés à des marqueurs de sélection comme URA et TRP - sous forme de plasmide dans des bactéries puis, après coupure enzymatique, liés à de grands fragments d'ADN (100-1 000 kilobases) avant introduction dans des protoplastes de levure Saccharomyces cerevisiae.
