Les cerveaux des individus atteints par la maladie de Parkinson contiennent des dépôts protéiniques, connus sous le nom de corps de Lewy, analogues aux plaques amyloïdes de la maladie d'Alzheimer. Ces dépôts contiennent des fibres composées de la protéine alpha-synucléine. Il y a quelques cas rares de maladie de Parkinson héréditaire et ces individus présentent une mutation dans le gène de l'alpha-synucléine. Normalement la protéine alpha-synucléine possède une conformation très flexible, non structurée.
