Many enzymes require coenzymes to exhibit their catalytic activities. Generally, the binding force between apoenzyme and coenzyme is relatively weak, and reversible separation to give the protein moiety (apoenzyme) and coenzyme can occur easily. Chez les enzymes de type hétéroprotéique, la fixation du substrat se fait sur la partie protéique, l'apoenzyme, tandis que la catalyse de la réaction est le fait de la partie non protéique, le groupement prosthétique ou le cosubstrat. La partie «apoenzyme» de la molécule enzymatique qui porte le site de fixation du substrat est donc d'abord responsable de la spécificité quant au substrat de l'enzyme.
