Les benzodiazépines agissent au niveau du système nerveux central sur des récepteurs spécifiques étroitement liés aux récepteurs du GABA [acide gamma-aminobutyrique] et au canal du chlore. Elles renforcent l'inhibition gabaergique centrale par leur action agoniste. Elles ont un effet anxiolytique, sédatif, anticonvulsivant, myorelaxant et elles induisent une amnésie antérograde. Celles actuellement utilisées en anesthésie ont un effet hypnotique important. Les trois benzodiazépinesadministrées par voie intraveineuse sont le diazépam (1959), le flunitrazépam (1973) et le midazolam (1976).
