A human clinical trial has been performed to assess the safety and toxicity of direct gene transfer in humans. This general approach also may provide a method for immunization against HIV or other antigens.

Dans le transfert de gène direct, in vivo, le gène est délivré directement par injection intracérébrale [dans le cerveau de l'animal sur qui est tenté l'expérience]. Le vecteur s'intègre ou non dans la cellule et l'effet thérapeutique est mis en évidence par divers tests biologiques. Dans l'approche ex vivo, des cellules provenant ou non de l'hôte sont mises en culture puis infectées par un vecteur recombiné. Les cellules génétiquement modifiées sont ensuite sélectionnées, caractérisées biochimiquement puis transplantées dans le SNC (système nerveux central).
