Cette carte bidimensionnelle est obtenue en effectuant une électrophorèse d'un mélange peptidique selon une direction de la feuille de papier, en séchant le papier, puis en effectuant une séparation chromatographique des peptides dans l'autre direction. Aucun des deux procédés ne permet à lui seul la séparation des peptides, mais la combinaison séquentielle des deux est très efficace pour séparer les peptides contenus dans un mélange complexe.
