The gel-clot technique (Method A and B) allows detecting or quantification of endotoxin and is based on clotting of the lysate in the presence of endotoxins. The concentration of endotoxins required to cause the lysate to clot under standard condition is the labelled lysate sensitivity.

MÉTHODOLOGIE : La technique de gélification en présence d'un lysat de Limolus polyphemus (LAL) a été utilisée pour déterminer les taux d'endotoxines de tous les vaccins.

OBS

L'addition d'une solution contenant des endotoxines à une solution de lysat provoque une turbidité, une précipitation ou une gélification du mélange. En effet, les cellules de l'hémolymphe [sang des arthropodes] d'un crabe, le Limulus polyphémus ont la particularité de se gélifier en présence de quantité même très faible d'endotoxines.
