Multiplication à l'échelle cellulaire. La pénétration se fait par fusion-lyse ou par pinocytose. La capside est désagrégée dans le cytoplasme et l'ADN viral passe dans le noyau. Son expression aboutit à la synthèse successive de trois lots de protéines : les protéines très précoces (immediate early), codées par les gènes [alpha] et culminant 3 heures après infection; les protéines précoces (early, 6 heures après infection) codées par les gènes y.
