C'est pour répondre à ce problème que la technique dite de la méga-amorce a été mise au point. Dans un premier temps, la matrice d'ADN parental est amplifiée par une PCR utilisant comme amorces un oligonucléotide qui porte la mutation désirée, et un deuxième oligonucléotide situé à proximité d'un site de restriction X. Le produit de cette PCR est appelé la méga-amorce : il correspond à une partie de la séquence du gène, joignant la mutation et le site X.
