Decay accelerating factor (DAF) and CR1 inhibit the association between C3b and B, and promote dissociation of the C3bBb complex. CR1 and membrane cofactor protein (MCP) promote Factor I mediated cleavage of C3b. These molecules control the interactions between C4b and C2 similarly.

Les «membranes cofactor proteins» (MCP) ou gp 45,70 : ces protéines constituent un nouveau groupe de protéines régulatrices fixant le C3b, elles sont présentes sur un grand nombre de cellules, mais, à la différence du DAF, elles sont absentes des hématies. Elles fixent le C3b et le C4b, ont une fonction cofacteur du I, favorisent l'inactivation du C3b et du C4b, sans toutefois dissocier les convertases. Elles jouent donc un rôle complémentaire à celui du DAF.
