In broad outline the plus-and-minus method calls for synthesizing pieces of DNA that are complementary to various regions of the DNA being sequenced. Because of the way the synthesis is manipulated each piece ends either with or just before a particular, identifiable nucleotide (A, G, T or C). By performing the synthesis to produce a large number of pieces, each one ending with a successive nucleotide in the sequence, it is possible to establish the sequence of the entire DNA molecule.

Dans ses grandes lignes, la méthode du «plus-moins» consiste en la synthèse de fragments d'ADN, complémentaires des zones de l'ADN dont on veut établir la séquence. La synthèse est orientée de telle façon que chaque fragment se termine par un nucléotide particulier que l'on peut identifier (A, G, T ou C) ou par le nucléotide qui le précède. En faisant la synthèse d'un grand nombre de fragments dont chacun se termine par un des nucléotides successifs de la séquence, il est possible d'établir la séquence entière de la molécule d'ADN.
