La technique microscopie à lumière polarisée est basée sur la propriété que possèdent des composants anisotropes ou biréfringents d'un échantillon à modifier la phase de la lumière polarisée. Les substances ayant une structure moléculaire cristalline ou laminaire, ou bien contenant des arrangements réguliers de bâtonnets submicroscopiques peuvent modifier de façon différentielle la vitesse de deux rayons de lumière polarisée vibrant dans des plans perpendiculaires. Cela produit un phénomène de biréfringence ressortant en blanc su fond noir.
