Pour que la recombinase puisse remplir sa fonction, il faut que la structure de la chromatine soit dans un état tel que l'enzyme puisse accéder aux signaux de reconnaissance. Selon Yancopoulos et al., cette condition est remplie lorsque des portions de chromatine (du même chromosome ou de deux chromosomes) sont transcriptionnellement actives au moment de l'événement de recombinaison.
