La chromatographie par formation de paires d'ions est, d'un point de vue formel, une technique permettant de faire de l'échange d'ions en utilisant des supports destinés à la chromatographie de partage. La chromatographie par formation de paires d'ions sur supports non polaires s'est surtout développée (reversed phase ion pair chromatography). Le soluté ionisé présent dans la phase mobile hydro-organique forme, avec un ion de signe contraire appelé contre-ion, une paire d'ions qui est retenue par la phase stationnaire organique.
