Sildenafil, ... was discovered through a rational drug design programme by the pharmaceutical company Pfizer. It is a potent selective inhibitor of the enzyme phosphodiesterase (PDE-5), which destroys cyclic guanosine monophosphate (cGMP), itself a dilator of blood vessels in the body.
