L'homogénat obtenu à l'homogénéiseur de Potter est centrifugé 10 minutes à 700 g. Le sédiment contient les noyaux, le surnageant, comprenant le cytoplasme, est transvasé dans un autre tube, centrifugé 10 minutes à 10 000 g, ce qui sédimente les mitochondries et les lysosomes. Le surnageant post mitochondrial est soumis à ultracentrifugation pendant 1 heure à la vitesse la plus grande possible, donnant une accélération d'au moins 100 000 g. On sédimente ainsi les microsomes dont la partie insoluble dans le désoxycholate (DOC) comprend les ribosomes. Le surnageant comprend le cytosol.

The two definitions are complementary not contradictory. "Supernatant" refers to the fact that the liquid is found above a precipitate be it in a sedimentation tank or a sludge digestion tank.
