Certaines mutations ne se manifestent qu'à une température relativement élevée et non à une température basse ou moyenne. On les qualifie de thermo sensibles. Elles correspondent souvent à une fragilité particulière de la protéine mutée vis-à-vis d'une élévation de température, fragilité qui ne se manifeste pas à une température plus basse, ce qui ne permet pas alors de déceler la mutation.
