Chronic wasting disease (CWD) is a progressive, fatal nervous system disease that affects animals which are all part of the deer family[, and] known as cervids. It is a transmissible spongiform encephalopathy, or prion disease. It is contagious amongst cervids, like scrapie in sheep. No virus has ever been causally associated with CJD [Creutzfeldt-Jacob disease]. If such a virus exists its properties are very unusual. It must be ubiquitous but normally non pathogenic. PrP [prion protein] would be essential for its infectivity e.g. perhaps serving as a receptor. Prion strains exist (more than 20 strains have been identified in mice infected with scrapie prions and several strains from humans). These strains breed true in inbred mice and differ in their incubation times and lesion profiles. This observation and the reluctance to accept because of precedence the existence of a replicating agent lacking a nucleic acid genome has led to the virino hypothesis. It proposes that a small nucleic acid in association eith PrP is the infectious agent. Scrapie, the prototype of a group of diseases which have the unique property of being both hereditary and infectious, is also exceptional in that it fails to evoke an immune response. Purification of crude scrapie preparations showed a strong association of infectivity with a membrane protein ('PrPSC'); but a protein with the same amino acid sequence ('PrPC') was subsequently also found in normal mammalian nervous tissue. Prions mediate the pathogenesis of certain neurodegenerative diseases, including bovine spongiform encephalopathy of cattle and Creutzfeldt-Jakob disease of humans. The prion particle consists mainly - if not entirely - of PrPSc, a posttranslationally modified isoform of the cellular host-encoded prion protein (PrPc).
