Pendant longtemps on n'avait pas su provoquer la "capacitation" du spermatozoïde, c'est-à-dire les transformations qui le rendent capable de féconder l'ovule. Cette capacitation semblait ne pouvoir se réaliser que par le passage du spermatozoïde dans l'utérus.
