À la surface interne du cytotrophoblaste et provenant de lui, naissent des cellules mésenchymateuses dont les plus internes forment une couche unicellulaire (membrane de Heuser ou membrane exocoelomique) s'unissant aux bords du disque entoblastique et constituant la limite d'une sphère creuse : le lécithocèle primaire (ou vésicule vitelline primitive ou cavité exocoelomique). Le mésenchyme extra-embryonnaire, qui a continué de proliférer, se creuse de cavités qui, par confluence, vont former la cavité choriale (ou coelome extra-embryonnaire).
