Tolerance that develops over time is termed acquired tolerance. Three classes of mechanism govern the development of acquired tolerance: pharmacokinetic, pharmacodynamic, and learned. Pharmacokinetic tolerance develops when the ability to metabolize or excrete the drug increases over time.
