La rupture de cellule peut s'effectuer par broyage au mixer, autolyse par du toluène et parfois par pression accompagnée ou non d'un léger chauffage (cas de la zymase), par traitement à l'acétone et à l'éther; si toutes ces méthodes sont insuffisantes, on peut faire agir les ultra-sons.
