La technique des plages d'hémolyse consiste à introduire des cellules lymphoïdes et une population dense d'érythrocytes xénogéniques dans un milieu gélifié. Certaines cellules lymphoïdes libèrent des anticorps hémolytiques qui diffusent autour d'elles et entraînent la lyse des hématies de leur voisinage, en formant une plage de lyse (on parle en anglais de «plaque forming cells» ou «PFC») quand on ajoute du complément.
