Classiquement le terme de stimulant respiratoire, tout comme celui d'analeptique, était réservé aux substances activant la commande ventilatoire. Ce sens restrictif était la conséquence d'un mode d'action reconnu : une neurostimulation induisant une hyperventilation. Les molécules actuelles semblent avoir, par opposition, un effet beaucoup plus diversifié. L'appareil respiratoire ayant pour rôle de régler les échanges gazeux, toute substance capable d'activer ces échanges peut prétendre aujourd'hui à être définie comme un stimulant respiratoire.
