Cette lignée devient ainsi «transcomplémentaire» pour le génome viral modifié et sera appelée «lignée d'encapsidation» (c'est-à-dire que le virus y trouvera le complément de son génome tronqué). Elle sera utilisée pour assurer la production du vecteur, qui aura donc à sa disposition les protéines nécessaires à sa réplication, que son génome, amputé, n'est plus capable de traduire.
