AC medium is a good primary isolation medium possessing unique growth promoting properties for both aerobic and anaerobic microorganisms. It is recommended as a general culture medium for anaerobes, microaerophiles and aerobes. The medium contains an agar flux that retards air diffusion and when sealed in snap-cap tubes immediately after autoclaving, it maintains an oxygen gradient ranging from anaerobic at the bottom to aerobic at the top. C'est un milieu de culture microbiologique d'utilisation générale (non sélectif) servant à cultiver une large gamme de micro-organismes (bactéries aérobies, anaérobies ou micro-aérophiles). Il est empirique (on ne connaît pas sa composition avec précision puisqu'il n'est pas purement synthétique). Celui qui est fabriqué par les laboratoires Sigma-Aldrich contient des peptones, des extraits de bœuf, de levure et de malt, ainsi que du dextrose, de l'acide ascorbique et de l'eau distillée. Membrane filtration is the method of choice for the analysis of fecal coliforms in water. Samples to be tested are passed through a filter of particular pore size(generally 0. 45 micrometre). The microorganisms present in the water remain on the filter surface. When the filter is placed in a sterile Petri dish and saturated with an appropriate medium, growth of the desired organisms is encouraged, while that of other organisms is suppressed.... A 100 ml volume of a water sample is drawn through a membrane filter(0. 45 µm pore size) through the use of a vacuum pump. The filter is placed on a Petri dish containing M-FC agar and incubated for 24 hours at 44. 5 °C(112. 1 degrees F). Le terme «coliformes fécaux« ou «coliformes thermotolérants» [désigne] toutes les espèces bactériennes faisant partie de la famille des Enterobacteriaceae qui sont aérobies ou anaérobies facultatives, à Gram négatif, asporulées, en forme de bâtonnet et produisant des colonies bleues en moins de 24 heures à 44,5 °C sur une gélose m-FC contenant du lactose. La méthode de filtration sur membrane consiste à recueillir, identifier et dénombrer, à la surface d'une membrane filtrante stérile, les bactéries coliformes d'origine fécale dans un échantillon d'eau. Il s'agit de filtrer à travers une membrane de porosité de 0,45 mm un volume déterminé de l'échantillon, de déposer cette membrane sur un milieu de culture sélectif, la gélose m-FC, et d'incuber cette gélose à 44,5 ± 0,2 °C pendant 24 ± 2 heures. Dans ces conditions, les coliformes fécaux forment des colonies bleues, permettant ainsi de les énumérer et de les identifier de façon présomptive. La présence de coliformes fécaux est confirmée ensuite par une réaction positive au test de l'ONPG [test d'utilisation de o-nitrophényl-β-D-galactopyranoside ou de présence de l'enzyme β-galactosidase] et une réaction négative à l'épreuve de la cytochrome-oxydase. Une confirmation spécifique de E. coli demande aussi une réaction positive de l'épreuve du MUG [test d'utilisation de 4-méthylumbelliféryl-β-D-glucuronide]. A medium containing peptone, lactose, dipotassium phosphate, agar, sodium sulfite, basic fuchsin, and distilled water. A hydrophilic colloidal material used largely in food products as emulsifying, thickening and gelling agents. They readily absorb water, thus increasing viscosity and imparting smoothness and body texture to the product, even in concentrations of less than 1%. Natural types are plant exudates(gum arabic), seaweed extracts(agar), plant seed gums or mucilages(guar gum), cereal gums(starches), fermentation gums(dextran), and animal products(gelatin). Semisynthetic types are modified celluloses and modified starches. Completely synthetic types are also available, e. g., polyvinylpyrrolidone. Granulated agar is used as a solidification agent in the preparation of microbiological culture media. [It] is a water soluble colloidal extract obtained from certain species of marine red algae including Gelidium, Pterocladia, and Gracilaria. Impurities, debris, minerals, and pigment are reduced to specific levels during manufacture. Well-known organic gel-forming substances are gelatin, collagen, metallic soaps, and such carbohydrate polymers as starches, agar, dextran and pectin.
