Chronic wasting disease (CWD) is a progressive, fatal nervous system disease that affects animals which are all part of the deer family[, and] known as cervids. It is a transmissible spongiform encephalopathy, or prion disease. It is contagious amongst cervids, like scrapie in sheep. There has been no known transmission of CWD to humans, however, bovine spongiform encephalopathy (also known as mad cow disease) in cattle has been known to cause Creutzfeldt-Jakob disease in humans. 6, fiche 1, Anglais, - chronic%20wasting%20disease%20of%20cervids

Prion diseases are rare, fatal, degenerative brain disorders that are thought to occur worldwide in both humans and animals. ... Although there are several forms of human prion disease, the most common is Creutzfeldt-Jakob disease (CJD). ... Prion diseases that affect animals include bovine spongiform encephalopathy (BSE) in cattle, scrapie in sheep and goats, and chronic wasting disease (CWD) in deer, moose, elk, and caribou or reindeer. Although they are rare, these diseases have caused great public health concern because of their many difficult and unusual features. 7, fiche 2, Anglais, - transmissible%20subacute%20spongiform%20encephalopathy

No virus has ever been causally associated with CJD [Creutzfeldt-Jacob disease]. If such a virus exists its properties are very unusual. It must be ubiquitous but normally non pathogenic. PrP [prion protein] would be essential for its infectivity e.g. perhaps serving as a receptor. Prion strains exist (more than 20 strains have been identified in mice infected with scrapie prions and several strains from humans). These strains breed true in inbred mice and differ in their incubation times and lesion profiles. This observation and the reluctance to accept because of precedence the existence of a replicating agent lacking a nucleic acid genome has led to the virino hypothesis. It proposes that a small nucleic acid in association eith PrP is the infectious agent. It is not fully consistent with all of the data and no such nucleic acid sequence has ever been detected. 1, fiche 15, Anglais, - virino

The transmissible spongiform encephalopathies are characterized by the conversion of the protease-sensitive prion protein (PrPsen) into a protease-resistant isoform (PrPres) associated with the neuropathogenic process in vivo. Recently, PrPres has been shown to be capable of directly inducing the conversion of PrPsen to PrPres in a cell-free in vitro system. In the present experiments, various PrP peptides were studied for their ability to enhance or inhibit this cell-free conversion reaction. None of the synthetic peptides was able to confer protease-resistance to the labeled PrPsen molecules on their own. 1, fiche 22, Anglais, - protease%2Dsensitive%20prion%20protein

Scrapie, the prototype of a group of diseases which have the unique property of being both hereditary and infectious, is also exceptional in that it fails to evoke an immune response. Purification of crude scrapie preparations showed a strong association of infectivity with a membrane protein ('PrPSC'); but a protein with the same amino acid sequence ('PrPC') was subsequently also found in normal mammalian nervous tissue. It is postulated by some investigators that 'PrPSC' is itself the infectious agent, or the most important part thereof, but in papers making that proposal immunological aspects have not been addressed. Experimental evidence supporting the hypothesis of a membrane fragment as agent has likewise lately not been taken into account. A modified form of the membrane hypothesis could account for immunological as well as genetic aspects of these diseases. 1, fiche 23, Anglais, - PrPSC

Prions mediate the pathogenesis of certain neurodegenerative diseases, including bovine spongiform encephalopathy of cattle and Creutzfeldt-Jakob disease of humans. The prion particle consists mainly - if not entirely - of PrPSc, a posttranslationally modified isoform of the cellular host-encoded prion protein (PrPc). It has been suggested that additional cellular factors might be involved in the physiological function of PrPc and in the propagation of PrPSc. 1, fiche 24, Anglais, - cellular%20host%2Dencoded%20prion%20protein
