Sequential isolation of overlapping molecular clones in order to span large intervals on the chromosome.

OBS

The technique depends on isolating a small segment of DNA from one end of the first recombinant and using this piece of DNA as a probe to rescreen the phage or cosmid library in order to obtain a recombinant containing that piece of DNA and the next portion of the genome. The second recombinant is used to obtain a third, and so on, to yield a set of overlapping cloned segments.

On ne peut pas cloner directement de longues séquences d'ADN mais on peut pratiquer une série de clonages successifs de fragments d'ADN en allant d'une extrémité d'un chromosome à l'autre. Pour cela, on prépare une première sonde qu'on utilise pour séparer, dans la bibliothèque de restriction du chromosome, plusieurs séquences d'ADN-complémentaire qui se recouvrent partiellement. On choisit une de ces sondes pour répéter l'opération. Elles sont utilisées pour trouver de nouveaux fragments de restriction du chromosome et ainsi de suite. En clonant chaque fois le segment de restriction désigné par la sonde, et en utilisant des endonucléases de restriction variées, on parcourt le chromosome d'un bout à l'autre.

Consiste en generar fragmentos de restricción de tamaño grande (o clones BAC); después de sondearlos, se identifica un único punto de partida. Se sintetizan nuevas sondas complementarias a secuencias del mismo fragmento (clon BAC) contiguas al punto de partida, y se utilizan para identificar diferentes fragmentos de restricción (clones BAC) que solapan con el seleccionado como punto de partida.
