Les oligonucléotides anti-sens sont des segments d’ADN complémentaires d’une portion de l'ARN messager du VIH(L'ARN messager est «direct», car il code directement les protéines devant être synthétisées par le ribosome). On pense qu'ils se fixent à l'ARN messager viral et empêchent ainsi les ribosomes de traduire l'ARN messager en protéines virales. Cependant les oligonucléotides sont rapidement dégradés par les enzymes cellulaires. Pour protéger les oligonucléotides anti-sens, on peut les renforcer en substituant un atome d’oxygène par un atome de soufre sur les liaisons phosphate entre les nucléotides.
