À l'issue d'une mutagenèse, on dispose d'une collection d'allèles mutants. En général, ces allèles confèrent le même phénotype. La question se pose alors de savoir combien de gènes sont mis en jeu pour obtenir cette collection d'allèles. La génétique a développé des méthodes qui permettent de répondre à cette question. Il existe deux méthodes : la complémentation et la cartographie. Nous allons commencer dans ce chapitre par le test de complémentation. C'est en effet le plus rapide à mettre en œuvre et son application permet d'avoir une bonne idée du nombre de gènes. Le test consiste dans la fabrication d'une souche pour laquelle les allèles mutants sont associés deux à deux en configuration trans et en l'observation du phénotype. Soit pour un couple d'allèles mutants a et b.
