L'immunofluorescence directe implique la fixation de l'échantillon (cellule ou micro-organisme) contenant l'antigène d'intérêt, sur une lame. Des anticorps marqués à la fluorescéine sont alors ajoutés à la lame et incubés. La lame est lavée pour enlever les anticorps non fixés puis examinée au microscope à fluorescence pour rechercher une fluorescence jaune-verte. La disposition de la fluorescence indique la localisation de l'antigène.
