Chromatographie sur colonne. La méthode consiste à utiliser une colonne réalisée au moyen d'un tube de verre [...] rempli de matière adsorbante (alumine, silice amorphe, pâte de cellulose, etc.) et qui constitue la phase fixe. La solution contenant les substances à séparer est versée en haut de la colonne et s'écoule à travers celle-ci. En général, les substances contenues dans la solution sont adsorbées, à tel point que, pour les extraire, il faut laver la colonne adsorbante avec un solvant approprié (phase mobile) jusqu'à ce que les diverses fractions s'écoulent, l'une après l'autre, à la base de la colonne.
