This test for mutagenicity, now called the Ames test, has found wide but not yet official use as a test for carcinogenicity on the simple presumption that what can cause a mutation is overwhelmingly likely also to be able to cause cancer.

Lorsque Bruce Ames a ouvert la voie qui allait conduire aux tests actuels des cancérigènes potentiels, il a, parmi les diverses manifestations cellulaires consécutives aux lésions du DNA, choisi d'utiliser la mutagénèse des bactéries. Le tests d'Ames, aujourd'hui classique, combine l'usage d'une bactérie et celui d'un extrait de foie de rat. L'organisme qui sert de «cobaye» est une bactérie, Salmonella typhimurium, porteuse d'une mutation (his-) qui l'a rendue incapable de fabriquer l'une des enzymes nécessaires à la synthèse de l'acide aminé histidine, composant indispensable des protéines.

Prueba de Ames. Prueba de mutagenicidad en la cual una cepa de salmonella typhimurium mutante, caracterizada por su incapacidad para sintetizar histidina, se expone a un material que se va a ensayar, en un medio de cultivo que no contiene histidina. Si alguna de las bacterias recupera la capacidad de sintetizar la histidina y en consecuencia forma colonias, se comprueba que el material sometido a prueba tiene propiedades mutagénicas. Se pueden usar cepas de otras bacterias (por ejemplo: escherichia coli).
