Les faux positifs obtenus sont les clones pour lesquels seul l'oligonucléotide de sélection (c'est-à-dire celui qui permet de déléter le site de restriction) s'est hybridé. La situation idéale de mutagénèse serait donc celle pour laquelle le site de mutation est proche d'un site unique de restriction. Dans ce cas, un seul oligonucléotide portant à la fois la mutation à introduire et la délétion du site de restriction sera utilisé, éliminant de fait le problème des faux positifs. 1, record 3, French, - faux%20positif
