We therefore propose to undertake the identification, assembly and test of the structural elements necessary for the proper function and stability of a mammalian artificial chromosome(MAC). To achieve this goal, we will create a chimaera between an existing Yeast Artificial Chromosome(YAC) and selected human and other mammalian genomic sequences. Human artificial chromosomes(HAC) have been considered as a possible vector for human gene therapy. In our previous work a minichromosome(MC), isolated from a patient, was transformed with a selectable marker gene(neo) which was targeted into its centromere; the MC was then size reduced, by X-ray exposure, to be easily manipulated and further engineered. Scientists report in Wednesday's edition of the journal Nature Genetics that they have created the first functioning, artificial human chromosome. The new technology, developed by researchers from Case Western Reserve University and Athersys Inc., may eventually provide a way to cure inherited diseases by altering the cell' s genetic structure.
