The protein kinase PKR is a component of the interferon-induced anti-viral defense mechanism in mammalian cells. Upon activation by binding double-stranded RNA in infected cells, the kinase down-regulates the cellular translational apparatus, and thus impairs viral protein expression. To overcome the inhibitory effects of PKR, viruses have developed efficient methods to prevent the activation or function of the kinase. Récemment, Michael Lai et son équipe de l'université de Los Angeles ont décrit un mécanisme expliquant la plus grande résistance de certaines souches de VHC à l'interféron. Ces chercheurs travaillent sur une enzyme, la protéine kinase PKR, qui fait avorter les virus en inhibant leur synthèse protéique. Scrapie, the prototype of a group of diseases which have the unique property of being both hereditary and infectious, is also exceptional in that it fails to evoke an immune response. Purification of crude scrapie preparations showed a strong association of infectivity with a membrane protein(’PrPSC') ;but a protein with the same amino acid sequence(’PrPC') was subsequently also found in normal mammalian nervous tissue. It is postulated by some investigators that ’PrPSC' is itself the infectious agent, or the most important part thereof, but in papers making that proposal immunological aspects have not been addressed. Experimental evidence supporting the hypothesis of a membrane fragment as agent has likewise lately not been taken into account. Recently, several mesoderm-inducing factors(MIFs) have been discovered. These cause isolated Xenopus animal caps to form mesodermal cell types such as muscle, instead of their normal fate of epidermis. The MIFs fall into two classes. One comprises members of the fibroblast growth factor(FGF) family, and the other members of the transforming growth factor type beta(TGF-beta) family. Of the latter group, the most potent is XTC-MIF, a protein produced by Xenopus XTC cells.
