Perfluorooctane sulphonate (PFOS) ... belongs to a group of organic compounds, perfluorinated organic substances, which have the shared feature that all the hydrogen atoms, bound to the carbon chain, are exchanged for fluorine atoms. A reactive sulphonate group has been bound to one end of the carbon chain. Further chemical groups are bound to the sulphonate group so that various types of surface-active substances are formed. These substances may then in some cases be broken down to PFOS in nature. Altogether there are several hundred different compounds where PFOS forms part of the structure.

A large proportion of the PFOS produced forms part of polymers, where the final product may contain residues of fluorinated starting substances that have not or have only partially reacted. PFOS is used for surface treatment and as an impregnating agent in a number of products such as carpets, furniture, paper, textiles and leather. Other PFOS-like substances are used as surface-active substances e. g. in cleaning products.

EPA [Environmental Protection Agency] is proposing a supplemental significant new use rule (SNUR) under section 5(a)(2) of the Toxic Substances Control Act (TSCA) for the following 75 substances: Perfluorooctanesulfonic acid (PFOSH) and certain of its salts (PFOSS), perfluorooctanesulfonyl fluoride (POSF), certain higher and lower homologues of PFOSH and POSF, and certain other chemical substances, including polymers, that are derived from PFOSH and its homologues. These chemicals are collectively referred to as perfluoroalkyl sulfonates, or PFAS. ... In the original proposed SNUR, these chemicals were referred to collectively as perfluorooctylsulfonates, or PFOS, but commenters noted that this generic usage of the term PFOS was inconsistent with the use by the manufacturer, the Minnesota Mining and Manufacturing Company (3M), of PFOS to refer only to chemicals with an eight-carbon, or C8, chain length.

Figure 6. 1 illustrates a typical open disposal trench at the Barnwell facility. The trench floor is sloped slightly and covered with a layer of sand to facilitate collection of percolating water in a french drain. A number of pipes allow any water that may collect in the french drain to be removed and analyzed. Presently waste, primarily in boxes, drums and cask liners, is stacked neatly in the trench, starting at the high end and progressing toward the low end. High specific activity waste is usually placed at the bottom of the trench. A separate narrower shielded trench is used at Barnwell for waste that has very high dose rates at the package surface. As the level of the waste reaches the design level, 2 m below grade, a layer of dry soil is added and allowed to filter down between the waste containers. When a trench is filled, it is covered with a cap of at least 0. 6 m of compacted native clay, followed by at least 0. 9 m of soil.

Short stretches of RNA have been found attached to the 5’ ends of many newly synthesized DNA chains. The starting points for DNA replication may therefore be recognized not by a DNA polymerase, but by a form of RNA polymerase. Following binding of a starter RNA polymerase, a number of ribonucleotides are sequentially linked together until a stop signal is read. The enzyme then detaches leaving a short RNA chain still bound to its DNA template. This RNA chain then serves as a primer onto which DNA polymerase III adds deoxynucleotides. Removal of the RNA primer occurs later through the action of other enzyme(s) which can digest away the RNA component of RNA/DNA hybrids. One enzyme that has this capacity is DNA polymerase I, whose primary task may be to close the gaps between RNA-primed DNA fragments.

On a trouvé de courts fragments d'ARN attachés aux extrémités 5' de nombreuses chaînes d'ADN nouvellement synthétisées. Les points de départ de la réplication de l'ADN pourraient donc être reconnus non pas par une ADN polymérase mais par une forme d'ARN polymérase. Après l'attachement d'une ARN polymérase de démarrage, un certain nombre de ribonucléotides sont séquentiellement liés les uns aux autres jusqu'à lecture d'un signal stop. L'enzyme se détache alors, laissant une courte chaîne d'ARN toujours liée à sa matrice ADN. Cette chaîne d'ARN sert alors d'amorce à laquelle l'ADN polymérase III ajoute des désoxynucléotides. Cet ARN amorce est éliminé plus tard par l'action d'autres enzymes (ou d'une autre enzyme) qui sont capables de digérer la partie ARN de l'hybride ARN/ADN. Une enzyme qui possède cette aptitude est l'ADN polymérase I, dont le rôle principal consiste à combler les espaces entre les fragments d'ADN amorcés par des ARN.

Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced. Subsequently, the development of efficient in vitro packaging systems and in situ hybridization techniques eliminated the need to enrich the starting DNA for the sequences of interest.
