Les chromosomes métaphasiques humains présentent divers types de bandes selon le traitement auquel ils sont soumis (colorants, température, enzymes protéolytiques ou DNases). Les bandes G(iemsa) positives représentent environ 50 % des bandes chromosomiques, l'autre moitié étant constituée de bandes G(iemsa) négatives. A more detailed map of a large YAC [yeast artificial chromosome] insert can be produced by subcloning, a process in which fragments of the original insert are cloned into smaller-insert vectors. We therefore propose to undertake the identification, assembly and test of the structural elements necessary for the proper function and stability of a mammalian artificial chromosome(MAC). To achieve this goal, we will create a chimaera between an existing Yeast Artificial Chromosome(YAC) and selected human and other mammalian genomic sequences. Le principe de ce système consiste à munir de grands fragments d'ADN exogène (humain par exemple) de tous les éléments qui permettent leur propagation dans la levure [...].
