Messenger RNA(mRNA) is a single-stranded RNA molecule that is complementary to one of the DNA strands of a gene. The mRNA is an RNA version of the gene that leaves the cell nucleus and moves to the cytoplasm where proteins are made. A suppressor gene that allows the insertion of an amino acid into the growing polypeptide chain in response to a nonsense codon. In the absence of such a suppressor, premature chain termination occurs. A close link between tRNA gene redundancy and the overall amino acid composition of yeast proteins was also observed. Regression analysis values for individual protein coding sequences proved to be effective descriptors of the translational selective pressure operating on a particular gene. A significantly stronger co-adaptation between codon choice and tRNA gene copy number was observed in highly expressed genes. A computer program, which runs on MS-DOS personal computers, is described that assists in the design of synthetic genes coding for proteins. The goal of the program is the design of a gene which(i) contains as many unique restriction sites as possible and(ii) uses a specific codon usage. The gene designed according to the criteria above is(i) suitable for ’modular mutagenesis’ experiments and(ii) optimized for expression. The program ’reverse-translates’ protein sequences into degenerated DNA sequences, generates a map of potential restriction sites and locates sequence positions where unique restriction sites can be accommodated. The nucleic acid sequence is then ’refined’ according to a specific codon usage to remove any degeneration. Les suppresseur sont surtout connus chez des microorganismes et leur effet est de rétablir un phénotype plus ou moins normal aux souches qui portent certaines mutations. Il en existe 2 catégories dont le mode d'action est fondamentalement différent. La première correspond à des gènes qui ouvrent une voie métabolique nouvelle compensant plus ou moins la coupure établie dans les chaînes métaboliques par la première mutation. Ce type de suppresseurs est spécifique d'un gène déterminé et supprime toutes les mutations pouvant l'atteindre [suppresseur intragénique]. Les suppresseurs de la seconde catégorie sont, au contraire, spécifiques de certains types de mutations, quel que soit le gène qu'elles aient pu atteindre. Leur action se situe au niveau des mécanismes de la synthèse des polypeptides. Ils apportent certaines perturbations définies aux règles du transfert d'information qui va de l'ADN aux chaînes polypeptidiques et sont pour cette raison qualifiés parfois de suppresseurs informationnels. Ils correspondent à la formation de molécules d'ARN de transfert anormales qui sont capables de traduire les codons non sens et de permettre ainsi la synthèse de polypeptides fonctionnels par des cistrons mutés porteurs de tels codons [suppresseur extragénique]. 1, fiche 35, Français, - suppresseur
