[The] method, called bioluminescence resonance energy transfer(BRET), uses a bioluminescent luciferase that is genetically fused to one candidate protein, and a green fluorescent protein mutant fused to another protein of interest. Bioluminescence in Renilla is due to the luciferase-catalyzed oxidation of luciferin. Anderson and Cormier have isolated a membrane-bounded, subcellular particle containing luciferase, a luciferin-binding protein with its associated luciferin and a green fluorescent protein all of which are necessary for bioluminescence and its control. When subjected to hypotonic solutions of calcium, these particles produce a flash of green light identical to the in vivo luminescence of Renilla. Du Pont and Duke University collaborators have developed the Biolistic gene gun, a particle delivery system which employs microcarriers coated with genetic material to insert DNA into animal cells. The microcarriers are accelerated on macrocarriers in a stream of helium to fire the microcarriers into external tissue or, using surgery, into the livers of mice.
