Grow the bacteria in Burks nitrogen-free medium supplemented with 0. 05% yeast extract for 48 hrs. in shake flasks containing 1LTR medium use 1%(W/V) glucose, sterilised separately as the carbon source. Harvest the bacteria, break by ultrasonication or in the French pressure cell, ultracentrifuge and keep the supernatant from the lysate as enzyme. La séparation des têtes des flagelles par sonication aux ultra-sons nous a permis de déterminer que la P21b se localise sur le flagelle des spermatozoïdes alors que la P25b est associée à la tête.
