Genomic DNA was isolated from young wheat leaves using the DNeasy Plant Mini Kit. After disruption, lysis buffer AP1 was added to the tissue and the mixture was incubated at 65°C for 10 min. Le motif «boîte albumen» est présent dans la plupart des promoteurs des gènes de prolamines des triticées. Des protéines nucléaires d'albumen reconnaissent soit la partie A soit la partie B de cette séquence. La partie B contient un palindrome similaire au site de liaison des facteurs de transcription Fos et Jun. Ces protéines de mammifères de la classe bZIP transactivent le proGLUCR dans un système végétal via ce palindrome. L'ADNc encodant une protéine se liant à la «boîte albumen» a été isolé. Cette protéine, WHMG1, est une petite protéine chromosomique abondante présente dans l'albumen et les jeunes feuilles de blé. Il est souvent délicat de contrôler avec précision la position de l'objet étudié en raison de sa taille et de sa fragilité.
