In order to prove the strand separation hypothesis, methods had to be found for the physical separation of parental and daughter DNA molecules. This was first accomplished through use of heavy isotopes such as N15. Bacteria grown in a medium containing the heavy isotope N15 have denser DNA than bacteria grown under normal conditions with N14. Heavy DNA can be separated from light DNA by equilibrium centrifugation in concentrated solutions of heavy salts such as cesium chloride. If DNA replication involves strand separation, definite predictions can be made about the density of the DNA molecules found after various growth intervals in a light medium. After one generation of growth, all the DNA molecules should contain one heavy and one light strand and thus be of intermediate hybrid density. This result is exactly what is observed. Historiquement, trois mécanismes possibles de réplication du DNA ont été proposés : 1) la réplication conservative de DNA. Chacun des brins du DNA parental est répliqué, sans séparation des brins, en donnant du DNA d’origine et du DNA néoformé. 2) la réplication semi-conservatrice du DNA lourd. A la première génération, chaque double hélice contient un des brins d’origine parentale. 3) la réplication dispersive. Pendant la réplication, les chaînes parentales se fragmentent de place en place, et les segments néoformés se recombinent, dans le même brin, avec des segments d’origine parentale. Les turbines axiales regroupent les Kaplan, les bulbes et les hélices. Ce sont toutes des turbines à réaction. Elles se rencontrent dans les aménagements à forts débits et faible chutes(de 2 à 30 m). ("Unusual" means a base other than A, G, C, or U.) Many of the unusual bases differ from normal bases by the presence of one or more methyl (-CH3) groups. Most, if not all, of the methyl groups are enzymatically added after the nucleotides are linked together by 3'-5' phosphodiester linkages. Very probably, the other unusual bases also arise by the enzymatic modification of a preexisting polynucleotide. The function of the unusual bases is not yet clear. They are not limited to alanine tRNA but occur in varying proportions in all tRNA molecules. Our only hint of their role is the fact that several unusual bases cannot form conventional base pairs. Some of the unusual bases may thus have the function of disruption double-helical hairpin regions, thereby exposing free keto and amino groups which can then form secondary bonds. Depending upon the specific bases, the free groups may form secondary bonds to template RNA, to a ribosome, or to the enzyme needed to attach a specific amino acid to its specific tRNA molecule. Mushroom arm cutter. Another special type of arm was the mushroom arm ... This consisted of a standard jib with a vertical tree or turrett mounted near its end. The turret was fitted with teeth and rotated about its axis. It was driven by the cutting chain on the main jib. The mushroom device was designed to fit on to any standard-type chain coal-cutter and, like the curved jib, it allowed the operator to make a shear cut at the back of the web at the same time as the horizontal cut was being made. Le champignon, inspiré des vielles haveuses à barre, est un petit tambour court portant des couteaux disposés en hélice. Il est fixé verticalement sur l'axe de l'étoile du retour, qui, entraînée par la chaîne, entraîne à son tour le champignon. Généralement, celui-ci est au-dessus du bras. Il travaille ainsi mieux car les fines qu'il produit s’évacuent facilement. C'est ce processus de «recopiage» des brins qui constitue la réplication. Il est assuré par un appareil enzymatique très sophistiqué, le complexe de réplication, composé d’une quinzaine d’enzymes. Le complexe de réplication avance à grande vitesse le long des brins séparés de la double hélice parentale : il traite environ 4 000 nucléotides par seconde(rappelons que l'ADN de la bactérie Escherichia coli comprend cinq millions de nucléotides et que l'ADN humain en comprend mille fois plus).
