Les propriétés des liaisons peptidiques imposent certaines contraintes au repliement des protéines. Ainsi, en raison de la répartition des électrons entre les atomes d'oxygène, de carbone et d'azote, la liaison peptidique n'est pas un axe de rotation libre : elle s'oppose à cette rotation. De ce fait, les quatre atomes des groupes -CO-NH- sont dans un même plan et la chaîne peptidique ne se replie quasiment que par des rotations autour des liaisons avec les carbones alpha.
