L'acide folique ou vitamine B9 est l'acide phtéroylglutamique apporté par l'alimentation sous forme de polyglutamates. Après hydrolyse dans les entérocytes en monoglutamates et absorption dans le jéjunum, ils traversent la barrière intestinale, sont réduits en tétrahydrofolates (THF) puis méthylés en N5 méthyl THF, forme active circulante et de stockage hépatique (avec circulation entérohépatique). La forme active tissulaire est l'acide tétrahydrofolique, THF, le méthyl libéré servant à la synthèse de la méthionine à partir de l'homocystéine.
