A procedure which uses a gel matrix combined with an enzyme or nucleic acid to obtain purification of biomolecules.

DEF

A technique of molecular separation in which molecules are attached to an insoluble (e.g., sepharose) matrix. Only those molecules which show affinity to the bound molecule (e.g., an antibody for its antigen) are retained. These trapped molecules can be subsequently eluted.

La méthode consiste à attacher de façon covalente sur un support solide un analogue stérique du substrat de l'enzyme (inhibiteur compétitif ou substrat modifié) en vue de former avec l'enzyme un complexe du type enzyme-substrat [...]. L'enzyme sera donc retardé lors du passage sur la colonne par rapport aux autres protéines qui, ne reconnaissant pas l'analogue, sortiront de la colonne plus rapidement que l'enzyme.
