La chambre de pression (dite aussi «bombe» ou chambre de Scholander). Cette technique, entièrement différente, consiste à placer à l'intérieur d'une chambre de pression un échantillon végétal (jeune rameau feuillé, système racinaire, mais le plus souvent une feuille) dès son prélèvement. L'extrémité du pétiole ou de la tige doit dépasser de quelques millimètres à l'extérieur. On injecte alors dans cette chambre un gaz sous pression (air azote) pour faire monter progressivement la pression dans la chambre.
