To define the sequences regulating TIMP expression, we generated transgenic mice that expressed the Escherichia coli beta-galactosidase gene under control of a 5’ region of the mouse TIMP gene containing-2158 to-58 by upstream of the initiator ATG.

L'introduction d'un gène étranger dans le patrimoine héréditaire d'un individu définit la transgénèse. Différentes techniques sont utilisées chez les mammifères pour y parvenir. Les deux premières ne permettent pas, à elles seules, de choisir l'endroit où le transgène s'intégrera. Il s'agit : (1) de la micro-injection dans l'œuf, qui permet d'introduire de grands fragments d'ADN exogène; (2) des rétrovirus recombinants défectifs qui permettent d'intégrer avec une fréquence élevée des fragments d'ADN dont la taille est cependant limitée par la place disponible dans la capside du rétrovirus.

Antithymocyte globulin(ATG) is a concentrated anti-human T-lymphocyte immunoglobulin preparation derived from rabbits after immunization with a T-lymphoblast cell line. ATG is an immunosuppressive product for the prevention and treatment of acute rejection following organ transplantation. ATG reduces the host immune response against tissue transplants or organ allografts.

CONT

Immunosuppressive therapy with antithymocyte globulin(ATG) plus cyclosporine is an effective alternative to stem-cell transplantation and improves blood counts and survival. Although horse ATG is the standard therapy, rabbit ATG is more potent in depleting peripheral-blood lymphocytes and is preferred in other clinical circumstances.

A baculovirus transfer vector was constructed using a cloned copy of the autography californica nuclear polyhedrosis virus(AcNPV) polyhedrin gene. The ATG translation initiation codon and 32 bases of the polyhedrin mRNA 5’ leader sequences were deleted and replaced with a cDNA clone of the A/PR/8/34 influenza virus haemagglutinin(HA) gene. The hybrid HA/polyhedrin gene was transferred to AcNPV and polyhedrin-negative recombinants identified which expressed HA activity.
